This is an antineoplastic that may cause inhibition of DNA synthesis by acting as a ribonucleotide reductase inhibitor. Indications: chronic myeloid leukaemia; myeloproliferative disorders; polycythemia vera; haemoglobinopathies (sickle-cells disease). Cautions: impaired renal function; pre-existing anaemia should be corrected before beginning therapy with hydroxyurea. Contra-indications: pregnancy & breast feeding. Side effect: myelosuppression, nausea, and skin reactions. Dose: 20-30mg/kg daily or 80mg/kg every third day. |
