This, an analogue of the natural purines, hypoxanthine and adenine, is an antineoplastic that act as an antimetabolite. It has also immunosuppressant properties. Indications: acute leukaemias and chronic myeloid leukaemia. Cautions: patients with impaired hepatic and renal function; hepatic function should be monitored periodically. Contra-indications: Pregnancy; breast-feeding. Side Effects: bone-marrow depression; gastrointestinal toxicities (less toxic than fluorouracil and methotrexate) Dose: Initially 2.5mg/kg daily. |
